GP + PD-1-Fudan University-biliary tract carcinoma refers to a combination chemo-immunotherapy regimen being investigated by Fudan University for the first-line treatment of advanced biliary tract carcinoma. This regimen, evaluated in the Phase 2 ZSAB-TOP trial, consists of the chemotherapy doublet gemcitabine and cisplatin (GP) combined with two immune checkpoint inhibitors: tislelizumab, a monoclonal antibody targeting programmed cell death protein 1 (PD-1), and ociperlimab, a monoclonal antibody targeting T-cell immunoreceptor with immunoglobulin and ITIM domains (TIGIT). The combination aims to leverage the cytotoxic effects of chemotherapy to release tumor antigens while simultaneously blocking multiple inhibitory pathways (PD-1 and TIGIT) to restore and enhance the anti-tumor immune response.
