Gemcitabine + etoposide is a combination chemotherapy regimen used primarily in the treatment of various cancers, including small-cell lung cancer (SCLC) and relapsed or refractory non-Hodgkin lymphoma (NHL). Gemcitabine is a nucleoside analog that acts as a nucleoside metabolic inhibitor. It is phosphorylated within cells to active metabolites that are incorporated into DNA, leading to masked chain termination and apoptosis of malignant cells. Etoposide is a topoisomerase II inhibitor that induces DNA strand breaks by stabilizing the enzyme-DNA complex, preventing religation of DNA strands during replication. The combination has demonstrated activity in SCLC and NHL with manageable toxicity profiles, providing palliative benefit especially for patients who may not tolerate more aggressive regimens[2][5][8].
