Drug intelligence / Profile preview
This is a combination regimen consisting of three drugs: - **Gemcitabine** is a nucleoside analog used as an antineoplastic agent. It inhibits DNA synthesis by incorporation into DNA and inhibition of ribonucleotide reductase, leading to cell death. It is primarily used in the treatment of various solid tumors, including pancreatic, ovarian, breast, and non-small cell lung cancers[1][2][5]. - **Irinotecan** is a topoisomerase I inhibitor that prevents DNA from unwinding by stabilizing the cleavable complex between topoisomerase I and DNA during replication. This leads to double-strand breaks and apoptosis in rapidly dividing cells. Irinotecan is mainly indicated for metastatic colorectal cancer but also used in other solid tumors such as pancreatic and ovarian cancers[2][3][5]. - **Allopurinol** is a xanthine oxidase inhibitor that reduces uric acid production. In oncology settings, it helps prevent hyperuricemia (tumor lysis syndrome) associated with rapid tumor breakdown during chemotherapy[7][10]. The combination of gemcitabine plus irinotecan has been studied for synergistic cytotoxic effects in several solid tumors (notably pancreatic and ovarian cancer), while allopurinol serves as supportive care to manage chemotherapy-induced hyperuricemia rather than directly contributing antitumor activity[2][5][7]. There are known pharmacokinetic interactions among these agents; co-administration may increase toxicity risk due to overlapping side effect profiles or altered drug metabolism[1][3].
