Overview

This is a combination regimen consisting of three agents: gemcitabine, irinotecan, and panitumumab. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, leading to cell death; it acts as an antimetabolite chemotherapeutic agent. Irinotecan is a topoisomerase I inhibitor that prevents DNA unwinding and replication in cancer cells. Panitumumab is a fully human monoclonal antibody targeting the epidermal growth factor receptor (EGFR), blocking ligand binding and downstream signaling pathways involved in tumor cell proliferation and survival. This combination has been studied primarily for advanced biliary tract cancers (such as cholangiocarcinoma) and has shown encouraging efficacy with manageable toxicity profiles[1][6]. The regimen leverages both cytotoxic chemotherapy (gemcitabine, irinotecan) and targeted therapy (panitumumab) to enhance antitumor activity.
