Overview

Gemcitabine + ivonescimab is an investigational combination therapy being evaluated for the treatment of advanced non-small cell lung cancer (NSCLC). Ivonescimab (also known as AK112 or SMT-112) is a humanized bispecific monoclonal antibody that concurrently targets Programmed Cell Death Protein 1 (PD-1) and Vascular Endothelial Growth Factor (VEGF), designed to promote anti-tumor immune responses while inhibiting angiogenesis within the tumor microenvironment. Gemcitabine is a pyrimidine antimetabolite chemotherapy agent that inhibits DNA synthesis and has demonstrated potential immunostimulatory effects, such as increasing T-cell infiltration and reducing immunosuppressive cell populations. This combination is currently being studied in the Phase II ORIGIN2 trial (SAKK 18/25), sponsored by the Swiss Cancer Institute (SAKK), specifically for patients whose disease has progressed following prior chemoimmunotherapy.
