Gemcitabine + oxaliplatin is a combination chemotherapy regimen widely used for the treatment of various advanced solid tumors, including non-small cell lung cancer, gallbladder cancer, urothelial carcinoma, and others. Gemcitabine is a synthetic pyrimidine nucleoside analog that inhibits DNA synthesis by being incorporated into DNA during replication and causing chain termination. Oxaliplatin is a third-generation platinum-based compound that forms crosslinks with DNA, inhibiting its replication and transcription and inducing apoptosis in tumor cells. The two drugs have distinct mechanisms of action but are both broad-spectrum cytotoxic agents; their toxicities do not overlap significantly, making the combination synergistic for antitumor activity[2][1][5]. This regimen is administered intravenously.
