Overview

gemcitabine + oxaliplatin + radiotherapy is a combination chemo-radiotherapy regimen evaluated for the treatment of locally advanced, unresectable pancreatic adenocarcinoma and distal cholangiocarcinoma. The protocol, studied by University Hospital, Ghent, involves the administration of gemcitabine (a nucleoside analog that inhibits DNA synthesis and ribonucleotide reductase) and oxaliplatin (a platinum-based agent that forms DNA cross-links) in conjunction with ionizing radiation. This multi-modal approach leverages the radiosensitizing properties of gemcitabine to enhance the efficacy of radiotherapy while providing systemic cytotoxic activity through the GemOx doublet. The regimen was assessed in a Phase 1/2 clinical trial to determine the feasibility and recommended dose of oxaliplatin when combined with fixed-dose gemcitabine and radiotherapy.
