Overview

This is a combination chemotherapy regimen consisting of three agents: - **Gemcitabine**: a nucleoside analog that inhibits DNA synthesis, used as an antimetabolite chemotherapeutic. - **Platinum compound** (most commonly cisplatin): a DNA crosslinking agent that induces apoptosis in rapidly dividing cells. - **Erlotinib**: an oral small-molecule inhibitor of the epidermal growth factor receptor (EGFR) tyrosine kinase. The combination has been studied primarily for advanced pancreatic cancer and non-small cell lung cancer. The rationale is to combine cytotoxic chemotherapy with targeted inhibition of EGFR signaling to enhance antitumor effects. However, clinical trials have shown limited additional benefit over doublet regimens and increased toxicity[1][2]. Erlotinib plus gemcitabine is approved for advanced pancreatic cancer; adding platinum increases toxicity without clear survival advantage[1][2].
