Overview

This combination therapy regimen consists of the chemotherapy drug gemcitabine and fractionated stereotactic body radiotherapy (SBRT), often delivered via the CyberKnife system. Gemcitabine is a pyrimidine antimetabolite that acts as a nucleoside analog, inhibiting DNA synthesis and inducing apoptosis in rapidly dividing tumor cells. SBRT is a highly precise form of external beam radiation therapy that delivers high doses of ionizing radiation to a localized tumor volume over a small number of fractions, causing lethal DNA double-strand breaks while minimizing damage to adjacent healthy tissues. This specific regimen has been evaluated by Stanford University in Phase II clinical trials for patients with locally advanced, unresectable pancreatic adenocarcinoma, aiming to improve local tumor control and potentially downstage tumors for surgical resection.
