Gemcitabine + vinorelbine is a combination chemotherapy regimen consisting of two small molecule cytotoxic agents, gemcitabine and vinorelbine. Gemcitabine is a nucleoside analog that inhibits DNA synthesis by incorporating into DNA during replication, leading to apoptosis in rapidly dividing cells. Vinorelbine is a semi-synthetic vinca alkaloid that disrupts microtubule assembly, inhibiting mitosis and cell division. This combination has been studied primarily for the treatment of advanced or metastatic non-small cell lung cancer (NSCLC), especially as second-line therapy after platinum-based regimens, as well as in relapsed or refractory aggressive lymphomas such as diffuse large B-cell lymphoma (DLBCL). The regimen has demonstrated variable efficacy across studies; some report modest response rates with acceptable toxicity profiles while others note increased toxicity without significant survival benefit compared to monotherapy or platinum-based combinations[1][2][3][4]. Administration is typically intravenous on days 1 and 8 of a 21-day cycle[6].
