Overview

Gemcitabine + vinorelbine + prednisolone (often abbreviated as GVP) is a combination chemotherapy regimen used primarily for the treatment of relapsed or refractory lymphomas, including both Hodgkin's lymphoma and aggressive non-Hodgkin's lymphomas. - **Gemcitabine** is a nucleoside analog that inhibits DNA synthesis by incorporating into DNA and causing chain termination, leading to apoptosis in rapidly dividing cells. - **Vinorelbine** is a semi-synthetic vinca alkaloid that disrupts microtubule formation during mitosis, inhibiting cell division. - **Prednisolone** is a synthetic glucocorticoid with anti-inflammatory and immunosuppressive properties; in oncology regimens, it induces apoptosis in certain malignant lymphoid cells. This combination has been evaluated in phase II studies for patients who are not candidates for platinum-based or high-dose chemotherapy. The regimen has demonstrated substantial activity with manageable toxicity profiles but dose-limiting hematological toxicities such as leucopenia and thrombocytopenia[1][2][3]. It is generally administered intravenously on days 1 and 8 of each cycle (gemcitabine/vinorelbine), with oral prednisolone given daily over several days per cycle.
