Gemcitabine elaidate + cisplatin is an investigational combination therapy in which gemcitabine elaidate (also known as CO-1.01), a fatty acid derivative of gemcitabine, is paired with cisplatin for the treatment of advanced solid tumors, particularly stage IIIb/IV non-small-cell lung cancer (NSCLC). Gemcitabine elaidate circumvents resistance associated with low tumor expression of the membrane transporter hENT1, entering cells independently of hENT1, which may render it more effective in certain patient populations compared to gemcitabine. Cisplatin is a platinum-based chemotherapy agent that causes DNA crosslinking and leads to apoptosis in rapidly dividing cells. The rationale for the combination is synergistic cytotoxicity, exploiting different mechanisms of action to target cancer cell proliferation and survival.
