Overview

Gemcitabine PLGA microparticles is an experimental anticancer formulation developed by researchers at the University of Castilla-La Mancha and the Iberian Centre for Research in Energy Storage. This drug delivery system consists of the small molecule chemotherapy gemcitabine encapsulated within poly(lactic-co-glycolic acid) (PLGA) microparticles. The formulation is synthesized using a double emulsion solvent evaporation method (water-in-oil-in-water) and is notable for being the first to employ ethyl lactate, an FDA-approved green solvent, in this specific synthesis process. The microparticles are designed to provide controlled release of gemcitabine, thereby improving its bioavailability and biodistribution for the treatment of various cancers, with a particular focus on pancreatic cancer. Gemcitabine itself acts as a pyrimidine antimetabolite that inhibits DNA synthesis by targeting ribonucleoside-diphosphate reductase and incorporating into DNA strands to induce chain termination.
