Overview

Gemeprost is a synthetic analogue of prostaglandin E1 (PGE1) used primarily in obstetrics and gynecology. It acts as an agonist at prostaglandin E receptors to induce cervical ripening and uterine contractions. Gemeprost is indicated for preoperative cervical dilation prior to transcervical intrauterine procedures and for therapeutic termination of pregnancy in the first and second trimesters—often in combination with mifepristone. The drug is administered as a vaginal suppository. Its mechanism involves binding to prostaglandin E2 receptor subtypes EP2 and EP3 on myometrial tissue, leading to increased uterine contractility and softening of the cervix through collagenase activation[1][2][6]. Gemeprost is not FDA-approved but has been marketed under brand names such as Cervagem (notably in Europe) and Preglandin (Japan)[2][5][7].
