Overview

Gemtuzumab ozogamicin is an antibody-drug conjugate (ADC) composed of a humanized IgG4 kappa monoclonal antibody (gemtuzumab) covalently linked to the cytotoxic agent calicheamicin. The antibody specifically targets the myeloid cell surface antigen CD33, which is expressed on leukemic myeloblasts in most patients with acute myeloid leukemia (AML). Upon binding to CD33-positive cells, the complex is internalized and calicheamicin is released inside the cell, where it induces double-stranded DNA breaks leading to cell cycle arrest and apoptosis. Gemtuzumab ozogamicin is primarily indicated for the treatment of newly diagnosed or relapsed/refractory CD33-positive AML in adults and children[1][2][3][4][5][6][8].
