Overview

Gemtuzumab ozogamicin + cytarabine is a combination therapy used in the treatment of acute myeloid leukemia (AML), particularly in relapsed, refractory, or newly diagnosed cases with favorable or intermediate risk. **Gemtuzumab ozogamicin** is an antibody-drug conjugate that targets CD33-positive leukemia cells using a humanized monoclonal antibody linked to the cytotoxic agent calicheamicin, inducing DNA double-strand breaks and triggering apoptosis. **Cytarabine** is a nucleoside analog that inhibits DNA synthesis, primarily targeting rapidly dividing cells. The combination exploits the selective targeting of CD33-positive cells by gemtuzumab ozogamicin as well as the broad antineoplastic activity of cytarabine. This regimen has been evaluated as induction and consolidation therapy in both relapsed/refractory AML and in patients in first complete remission and may be especially effective in patients with NPM1-mutated AML[1][2][3][5].
