Gemtuzumab ozogamicin + cytarabine + daunorubicin is a combination regimen used primarily for the treatment of acute myeloid leukaemia (AML), particularly in newly diagnosed patients whose leukaemia cells express CD33. Gemtuzumab ozogamicin is an antibody-drug conjugate consisting of a monoclonal antibody targeting CD33 linked to the cytotoxic agent calicheamicin. It binds to CD33 on AML cells, delivering the chemotherapy directly into these cells where calicheamicin induces DNA breaks and cell death. Cytarabine is an antimetabolite chemotherapy that inhibits DNA synthesis by acting as a nucleoside analog, while daunorubicin is an anthracycline antibiotic that intercalates into DNA and inhibits topoisomerase II, leading to inhibition of DNA replication and repair. This combination has demonstrated improved event-free survival compared to standard induction regimens in clinical trials for AML[2][3][5][6][7][8].
