GEN-001 + pembrolizumab + mFOLFOX is a combination investigational drug regimen consisting of GEN-001, an oral microbiome therapeutic, pembrolizumab (a PD-1 immune checkpoint inhibitor), and mFOLFOX (modified FOLFOX, a chemotherapy regimen of oxaliplatin, folinic acid (leucovorin), and fluorouracil). GEN-001 is designed to modulate the gut microbiome to enhance antitumor immunity. Pembrolizumab blocks the PD-1 receptor on T cells, enhancing immune system activity against cancer. mFOLFOX is a standard chemotherapeutic regimen used primarily for colorectal and other gastrointestinal cancers, working by damaging DNA or interfering with cell division, especially in rapidly dividing cancer cells[1][3][4][5]. The combination is being studied in advanced cancers to harness synergistic cytotoxic, immunomodulatory, and microbiome-mediated effects.
