GEN-003 + Matrix-M2 is an investigational therapeutic vaccine developed by Genocea Biosciences for the treatment of genital herpes (HSV-2 infection). It is a protein subunit vaccine consisting of two recombinant T cell antigens: an internal fragment of the immediate early protein ICP4 (GB208) and glycoprotein D (gD2; GB217). These antigens are formulated with Matrix-M2, a saponin-based immune-stimulating adjuvant licensed from Novavax. The vaccine is designed as a first-in-class T cell-directed immunotherapy intended to elicit both T cell and B cell (antibody) immune responses against HSV-2. In Phase 2 clinical trials, the vaccine demonstrated significant reductions in viral shedding and lesion frequency. Although Genocea discontinued internal development in 2017, the program's assets were later optioned to Shionogi & Co. for potential global development and commercialization.
