Overview

GEN-004 is a T cell-directed, protein subunit pneumococcal vaccine developed by Genocea Biosciences. It was designed as a potential universal pneumococcal vaccine to provide protection against all strains of *Streptococcus pneumoniae* by targeting three conserved protein antigens (SP0148, SP1912, and SP2108). These antigens were identified using the ATLAS platform and are associated with protective TH17 T cell responses in humans. Unlike existing vaccines that primarily induce B cell (antibody) responses against selected serotypes, GEN-004 aims to stimulate T cell-mediated immunity to reduce or prevent nasopharyngeal colonization—the first step in infection—across all known pneumococcal strains. The product was tested in phase 1 and phase 2 clinical trials for prevention of pneumococcal infections but development has been discontinued[1][4][5][6].
