GEN-011 + interleukin-2 is an investigational combination immunotherapy regimen developed by Genocea Biosciences for the treatment of advanced solid tumors. GEN-011 is an autologous, personalized neoantigen-targeted T-cell therapy comprised of CD4+ and CD8+ T cells (referred to as PLANET cells) derived from the patient's peripheral blood. The therapy utilizes the proprietary ATLAS platform to identify up to 30 optimal neoantigens that drive anti-tumor immune responses while specifically excluding 'Inhibigens'—antigens that may promote tumor progression or suppress immune activity. In the TiTAN-1 Phase 1/2a clinical trial, GEN-011 was administered following lymphodepletion and in combination with interleukin-2 (IL-2), a cytokine used to promote the expansion, survival, and persistence of the infused T cells. The program targeted multiple indications, including melanoma and various squamous cell carcinomas. However, following disappointing clinical results and financial challenges, Genocea Biosciences announced plans to wind down operations in 2022, leading to the termination of the GEN-011 development program.
