Overview

GEN-27 is a synthetic derivative of the soy-derived isoflavone genistein, specifically 7-O-(4-chlorobenzyl)genistein, developed to enhance the anti-inflammatory and anti-tumor properties of the parent compound. It is primarily investigated as a potential chemopreventive agent for inflammation-associated colorectal cancer. GEN-27 exerts its effects by modulating the NF-κB signaling pathway; it inhibits the phosphorylation of IκB and IKKα/β, thereby preventing the nuclear translocation of the NF-κB p65 subunit. This inhibition leads to a significant reduction in the secretion of proinflammatory cytokines, such as interleukin-6 (IL-6) and interleukin-1β (IL-1β), which are critical drivers of tumor cell proliferation in the intestinal microenvironment.
