Overview

GEN0101 is an inactivated oncolytic virus therapy derived from the hemagglutinating virus of Japan envelope (HVJ-E). It is designed for intratumoral or subcutaneous administration and exerts antitumor effects through two main mechanisms: direct cytotoxicity against tumor cells and activation of antitumor immunity. Mechanistically, upon administration, GEN0101 targets and binds to the cytosolic nucleic acid receptor retinoic acid-inducible gene I (RIG-I), leading to immune activation[2]. Clinical studies have shown that it increases natural killer (NK) cell activity and interferon-gamma levels in patients[1][5][7]. The drug has demonstrated safety and tolerability in phase I/II trials for advanced melanoma and castration-resistant prostate cancer. It was initially developed by GenomIdea as a novel immunostimulant for various cancers including melanoma, mesothelioma, and prostate cancer[3][4][8].
