GEN0101 + pembrolizumab

GEN0101 + pembrolizumab is a combination therapy under investigation for advanced melanoma and potentially other cancers. GEN0101 is a clinical-grade formulation of the hemagglutinating virus of Japan-envelope (HVJ-E), also known as inactivated Sendai virus, which induces direct tumor cell death and stimulates antitumor immunity by activating T cells and natural killer (NK) cells. Pembrolizumab is a humanized monoclonal antibody that targets programmed cell death protein 1 (PD-1), blocking its interaction with PD-L1/PD-L2 to enhance immune-mediated tumor destruction. The combination aims to synergistically boost both innate and adaptive immune responses against tumors. GEN0101 has shown safety, tolerability, increased NK cell activity, elevated interferon-gamma levels, and promising antitumor effects in early-phase trials for advanced melanoma[6][8]. Pembrolizumab is approved for multiple cancer types including melanoma, lung cancer, head and neck cancer, among others[4][5].
