Overview

GEN1042 + pembrolizumab + chemotherapy is an investigational combination therapy for advanced or metastatic solid tumors. GEN1042 (also known as DuoBody-CD40x4-1BB) is a bispecific antibody that engages both the **CD40 receptor** (expressed on antigen-presenting cells and tumor cells) and **4-1BB** (a co-stimulatory immune receptor on T cells), designed to activate tumor-directed immune responses. Pembrolizumab is a monoclonal antibody checkpoint inhibitor targeting the programmed cell death 1 (**PD-1**) receptor, blocking its interaction with PD-L1/PD-L2, and thus potentiating T-cell-mediated antitumor immunity. The chemotherapy component may include various agents (such as fluorouracil, cisplatin, gemcitabine, carboplatin, pemetrexed, or paclitaxel), aiming at direct cytotoxicity of tumor cells. This combination is being tested to determine dose, safety, pharmacokinetics, and efficacy against diverse advanced solid tumors including but not limited to melanoma, non-small cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), pancreatic ductal adenocarcinoma (PDAC), and colorectal cancer (CRC)[1][2][4].
