Overview

GEN1047 is a bispecific monoclonal antibody developed by Genmab for the treatment of malignant solid tumors. It is designed to simultaneously bind to CD3 on T cells and B7-H4 (V-set domain-containing T-cell activation inhibitor 1) on tumor cells. By engaging both targets, GEN1047 redirects and activates T cells to kill B7-H4–expressing tumor cells through T cell-mediated cytotoxicity. The drug was administered intravenously as a solution for infusion in clinical trials targeting advanced or metastatic breast cancer, endometrial cancer, ovarian cancer, and squamous non-small-cell lung cancer (NSCLC-SCC). Development of GEN1047 was discontinued in late 2024/early 2025 after Phase I/II studies due to insufficient differentiation from other therapies[1][3][4][5][6][8].
