Overview

GEN1053 (also known as BNT313 or HexaBody-CD27) is a novel monoclonal antibody designed to target CD27, a costimulatory receptor on T cells involved in immune activation. Developed using Genmab’s proprietary HexaBody technology, GEN1053 is engineered to form antibody hexamers upon binding its target on the cell membrane of T cells. This unique structure enables potent CD27 agonist activity independently of Fc gamma receptor (FcγR)-bearing cells, which are often scarce in tumors. The Fc domain of the antibody has been further modified to silence Fc-mediated effector functions and prevent T-cell depletion. The drug aims to (re)activate and enhance antitumor immunity by stimulating T-cell activation, proliferation, and differentiation. GEN1053 is being developed for the treatment of malignant solid tumors and is currently under investigation as both monotherapy and in combination with immunomodulators[1][2][4][7][8].
