GEN1055 is an investigational bispecific monoclonal antibody developed using Genmab's HexaBody technology. It targets OX40 (also known as CD134), a costimulatory receptor expressed on activated T cells. Unlike traditional OX40 agonists that require Fcγ receptor crosslinking for activity, GEN1055 is designed to be Fcγ receptor crosslinking-independent and uses the HexaBody format to enhance complement-dependent cytotoxicity (CDC) by promoting antibody hexamerization on the cell surface. This leads to superior C1q binding and enhanced immune activation against tumor cells[1][2][7]. The drug is being evaluated in clinical trials as monotherapy and in combination with pembrolizumab (an anti-PD-1 antibody) with or without chemotherapy for the treatment of malignant solid tumors[3][4][5]. The primary goals are to assess safety, determine optimal dosing regimens, and evaluate preliminary antitumor efficacy.
