Overview

GEN1059 (also known as BNT314) is an investigational bispecific antibody candidate being jointly developed by Genmab and BioNTech using the proprietary DuoBody technology platform. It is designed to enhance antitumor immune responses by simultaneously targeting and activating two key co-stimulatory pathways: CD40 and 4-1BB (CD137). By acting as a dual agonist, GEN1059 aims to stimulate antigen-presenting cells through CD40 and provide potent co-stimulatory signals to T cells via 4-1BB, thereby promoting T-cell activation, proliferation, and effector functions within the tumor microenvironment. This dual-targeting approach is intended to overcome the limitations of monospecific agonists and improve therapeutic outcomes in patients with advanced or metastatic solid tumors. The drug is currently undergoing evaluation in Phase 1/2 clinical trials to assess its safety and preliminary antitumor activity.
