GEN1079 is an investigational bispecific monoclonal antibody developed by Genmab using its proprietary DuoBody platform. It is designed as a T-cell engager that simultaneously binds to the CD3 epsilon subunit of the T-cell receptor complex and 5T4 (trophoblast glycoprotein), an oncofetal antigen frequently overexpressed in various advanced solid tumors but with limited expression in normal adult tissues. By bridging T cells and 5T4-positive tumor cells, GEN1079 facilitates the formation of an artificial immunological synapse, leading to T-cell activation and the subsequent release of cytotoxic granules to induce tumor cell lysis. The drug is currently being evaluated in first-in-human Phase 1 clinical trials to determine its safety, tolerability, and preliminary efficacy in patients with select advanced malignant solid tumors.
