GEN1106 is a human monoclonal antibody developed by Genmab that targets **CD39** (ectonucleoside triphosphate diphosphohydrolase 1). CD39 is an enzyme highly expressed on various immune cells and tumor cells within the tumor microenvironment, where it catalyzes the conversion of extracellular adenosine triphosphate (ATP) and adenosine diphosphate (ADP) into adenosine monophosphate (AMP). This enzymatic activity is the rate-limiting step in the generation of adenosine, a potent immunosuppressive molecule that inhibits T-cell and NK-cell activity. By blocking CD39, GEN1106 prevents the accumulation of adenosine and maintains high levels of pro-inflammatory ATP, thereby reversing immune suppression and enhancing anti-tumor immunity. It is currently being evaluated in Phase 1 clinical trials for the treatment of advanced solid tumors, including metastatic urothelial carcinoma.
