GEN1119 (HexaBody-CD27) is a humanized monoclonal antibody agonist targeting CD27, a co-stimulatory receptor in the tumor necrosis factor receptor (TNFR) superfamily. Developed by Genmab, GEN1119 utilizes the proprietary HexaBody technology, which introduces a specific mutation (E430G) in the Fc region to promote the formation of antibody hexamers upon binding to CD27 on the cell surface. This hexamerization facilitates the clustering of CD27 receptors, mimicking the natural ligand-induced activation more effectively than standard bivalent antibodies. The resulting potent co-stimulatory signal enhances T-cell proliferation, survival, and effector functions, aiming to overcome immunosuppression within the tumor microenvironment. GEN1119 is currently in Phase 1 clinical development for the treatment of advanced solid tumors.
