GEN3014 (HexaBody-CD38) is a next-generation, hexamerization-enhanced human IgG1 monoclonal antibody targeting CD38. It incorporates the E430G mutation in its Fc domain, which promotes efficient antibody hexamer formation upon binding to cell surfaces. This structural modification enhances complement-dependent cytotoxicity (CDC), leading to potent tumor cell killing. GEN3014 also induces other Fc-mediated effector functions such as antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), trogocytosis, and apoptosis after Fc-crosslinking. Additionally, it inhibits CD38 cyclase activity, potentially relieving immune suppression in the tumor microenvironment. Developed by Genmab, GEN3014 has been investigated primarily for relapsed or refractory multiple myeloma and other hematologic malignancies[1][3][8].
