Overview

GENA-111 is a human monoclonal antibody-drug conjugate (ADC) targeting the Lutheran/basal cell adhesion molecule (BCAM), also known as CD239. Developed by Genome & Company in collaboration with Debiopharm, the ADC consists of a human IgG4 (S228P) antibody conjugated to the microtubule inhibitor auristatin F (AF) via Debiopharm's proprietary Multilink technology. BCAM is a cell surface glycoprotein overexpressed in various epithelial malignancies, including breast and skin cancers, while showing limited expression in normal tissues. Upon binding to BCAM, the ADC is internalized, releasing the cytotoxic payload to induce cell cycle arrest and apoptosis. Preclinical studies have demonstrated significant anti-tumor activity and complete remissions in xenograft models of BCAM-positive cancers.
