GENA-119B31 is a humanized monoclonal antibody developed by Genome & Company that targets the amyloid precursor protein (APP). APP is a type I transmembrane protein expressed on T cells that acts as a novel immune checkpoint by interacting with contactin 4 (CNTN4) on tumor cells, thereby mediating immunosuppressive signals. GENA-119B31 effectively blocks the APP/CNTN4 interaction, demonstrating promising T cell-mediated cytotoxicity in vitro and significant anti-tumor activity in preclinical murine models of pancreatic cancer and liver metastasis. The drug is designed to overcome resistance to standard immune checkpoint inhibitors, such as anti-PD-1 therapy, particularly in patients whose tumor-infiltrating T cells exhibit elevated APP levels.
