Overview

GENA-120 is an antibody-drug conjugate (ADC) targeting integrin beta 4 (ITGB4), developed by Genome & Company. It consists of a humanized anti-ITGB4 antibody (GENA-120B17) conjugated to the topoisomerase I inhibitor payload exatecan via a cleavable hydrophilic linker (LinkerE). ITGB4 is a tumor-specific antigen identified through the GNOCLE™ platform, highly expressed in solid tumors like colorectal, head and neck, and esophageal cancers, while showing minimal expression in normal tissues. GENA-120 works by binding to ITGB4, internalizing into cancer cells, and releasing exatecan to induce cell death and bystander killing. Preclinical studies have demonstrated potent antitumor activity and a favorable safety profile.
