Drug intelligence / Profile preview
GENC-231 is a selective small-molecule inhibitor of NUAK family kinase 1 (NUAK1) under development by Genome and Company for the treatment of kidney fibrosis. NUAK1 is a key driver of tubulo-interstitial fibrosis, promoting myofibroblast activation and extracellular matrix (ECM) gene expression. By selectively targeting NUAK1, GENC-231 suppresses TGF-beta1-induced fibrotic markers and fibroblast contraction. In preclinical studies, GENC-231 has demonstrated anti-fibrotic efficacy in adenine-induced and unilateral ureteral obstruction (UUO) mouse models, both as a monotherapy and in combination with standard-of-care therapies such as dapagliflozin and losartan, resulting in improved renal function and reduced tissue injury.
