Cytovia Therapeutics is developing gene-edited induced pluripotent stem cell (iPSC)-derived natural killer (iNK) cells as an allogeneic, off-the-shelf cell therapy for the treatment of solid tumors, primarily hepatocellular carcinoma (HCC). Utilizing TALEN gene-editing technology, these cells are engineered with specific functional enhancements to overcome the limitations of conventional NK cell therapies. Key modifications include the knock-in of Interleukin-15 (IL-15) to promote cell persistence and metabolic fitness, and the knock-out of the Transforming Growth Factor-beta Receptor type 2 (TGFβR2) to render the cells resistant to the immunosuppressive effects of TGF-beta within the tumor microenvironment. This platform aims to provide a scalable and potent immunotherapy that can be used as a monotherapy or in combination with multi-specific antibodies to enhance tumor targeting.
