Overview

The generation 2.5 microRNA-22-3p antagomir is a peptide nucleic acid (PNA)-based antisense oligonucleotide developed by AptamiR Therapeutics for the treatment of metabolic diseases. It specifically targets and antagonizes miR-22-3p, a microRNA that plays a key role in regulating lipid metabolism, thermogenesis, and insulin sensitivity. To enhance delivery and potency, these compounds are conjugated to either a fatty acid (such as C32:6 dotriacontahexaenoic acid) or a short peptide (such as hexarelin), which facilitates active uptake into adipocytes via the FAT/CD36 (Fatty Acid Translocase) transporter. This targeted delivery strategy is designed to improve the safety profile and reduce the required dose compared to earlier generations. The drug is currently in IND-enabling preclinical development for obesity, type 2 diabetes, and metabolic dysfunction-associated steatotic liver disease (MASLD/MASH).
