Genetically attenuated Plasmodium falciparum Δmei2 sporozoites is a live-attenuated whole-parasite vaccine candidate developed by the Leiden University Medical Center (LUMC) for the prevention of falciparum malaria. The vaccine utilizes a CRISPR-Cas9 engineered strain of *Plasmodium falciparum* where the *mei2* gene, which encodes an evolutionarily conserved RNA-binding protein, has been deleted. This genetic modification causes the parasite to arrest late in its liver-stage development, preventing the transition to the symptomatic blood-stage of infection. By allowing the parasite to persist longer in the liver than earlier-generation genetically attenuated parasites (GAPs), PfΔmei2 aims to elicit a broader and more potent repertoire of T-cell and antibody responses against pre-erythrocytic antigens, potentially providing enhanced protective immunity.
