Genetically Defined Degrader MGx is a molecular glue degrader (MGD) program being developed by TRIANA Biomedicines for the treatment of various solid tumors. The program utilizes TRIANA's proprietary platform to identify small molecules that act as "molecular glues," inducing or stabilizing a novel interaction between an E3 ubiquitin ligase and a specific target protein associated with genetically defined vulnerabilities in cancer. This interaction leads to the polyubiquitination of the target protein and its subsequent degradation by the 26S proteasome. By targeting proteins that were previously considered "undruggable," this approach aims to provide new therapeutic options for patients with specific genetic drivers of cancer. As of early 2024, the program is in the hit-finding stage of preclinical development, with specific protein targets remaining undisclosed.
