Overview

This investigational autologous T cell therapy is being developed by the Roswell Park Comprehensive Cancer Center for the treatment of recurrent or refractory gynecologic malignancies, including ovarian, primary peritoneal, and fallopian tube cancers. The therapy involves the ex vivo genetic modification of a patient's own T lymphocytes to express an affinity-enhanced T-cell receptor (TCR) specific for the NY-ESO-1 (CTAG1B) cancer-testis antigen. NY-ESO-1 is a tumor-associated antigen frequently expressed in ovarian cancers but with highly restricted expression in normal adult tissues, making it a selective target for immunotherapy. In clinical evaluation, these engineered T cells are often administered following lymphodepleting chemotherapy and in combination with agents like decitabine, which may upregulate NY-ESO-1 expression on tumor cells to enhance therapeutic efficacy.
