Geneticin (G418) is an aminoglycoside antibiotic structurally similar to gentamicin B1 and produced by the bacterium Micromonospora rhodorangea. It acts by binding to the 80S ribosome in both prokaryotic and eukaryotic cells, inhibiting protein synthesis at the elongation step and resulting in cell death. Resistance to Geneticin is conferred by the neo gene from Tn5 encoding aminoglycoside 3'-phosphotransferase (APH(3')II), which inactivates the drug. Geneticin is widely used as a selective agent in laboratory research for selecting genetically engineered cells that have incorporated a resistance gene; it is not approved for clinical use or therapeutic applications[2][3][6].
