Drug intelligence / Profile preview
Geniposidic acid is a natural iridoid glucoside found in various medicinal plants, including *Eucommia ulmoides* (Tochu) and *Gardenia jasminoides*. It exhibits a wide range of pharmacological activities, including anti-inflammatory, antioxidant, hepatoprotective, antihypertensive, and antifibrotic effects. Mechanistically, geniposidic acid acts as an agonist of the glucagon-like peptide-1 receptor (GLP-1R), which stimulates the secretion of atrial natriuretic peptide (ANP) to lower blood pressure. It also functions as a farnesoid X receptor (FXR) modulator and sirtuin 6 (SIRT6) activator, regulating bile acid and cholesterol metabolism, which contributes to its protective effects against drug-induced liver injury (DILI) and metabolic dysfunction-associated steatotic liver disease (MASLD). Additionally, geniposidic acid has been shown to suppress hepatic stellate cell (HSC) activation via modulation of the AKT/GSK-3β signaling pathway, highlighting its potential as an antifibrotic therapeutic.
