**GeniusVac-Mel4** is an investigational, off-the-shelf allogeneic plasmacytoid dendritic cell line-based therapeutic cancer vaccine developed for HLA-A*02:01-positive metastatic melanoma. The product consists of irradiated plasmacytoid dendritic cells separately loaded with four HLA-A*02:01-restricted melanoma-antigen peptides derived from Melan-A, MAGE-A3, gp100, and tyrosinase. Following subcutaneous administration, the peptide-presenting dendritic cells are intended to prime and expand melanoma-antigen-specific cytotoxic CD8+ T-cell responses. In a completed first-in-human phase 1 dose-escalation study in nine patients with advanced melanoma, the vaccine was generally well tolerated and induced detectable expansion and memory differentiation of Melan-A-specific T cells in a subset of participants.
