GenSci133 (also known as GenSci-133 or GenSciP133) is a novel recombinant bifunctional fusion protein developed by GeneScience Pharmaceuticals and its affiliate North America Genescience for the treatment of osteoporosis. The molecule is designed with a dual-action mechanism, consisting of an anti-RANKL single-arm antibody fused to a parathyroid hormone (PTH) analogue. The anti-RANKL component binds to and inhibits Tumor Necrosis Factor Ligand Superfamily Member 11 (RANKL) to suppress bone resorption, while the PTH analogue component binds to and activates the Parathyroid Hormone 1 Receptor (PTH1R) to stimulate bone formation. This synergistic approach aims to achieve a net increase in bone mass. Developed using the proprietary LASR long-acting platform, GenSci133 is formulated as a subcutaneous injection intended for administration once every 3 to 6 months. In September 2026, the National Medical Products Administration (NMPA) of China approved the Investigational New Drug (IND) application for GenSci133, clearing it to enter Phase I clinical development.
