Overview

GenSci140 is a bispecific antibody-drug conjugate (ADC) targeting folate receptor alpha (FRα) for the treatment of advanced solid tumors, including platinum-resistant ovarian cancer. It simultaneously binds two different epitopes of FRα on tumor cells via a combination of Fab fragments and single-domain antibodies (VHH), leading to increased tumor cell binding and internalization. The ADC employs a cleavable linker to deliver a cytotoxic payload of a DNA topoisomerase I inhibitor (TOPOI). The released payload blocks DNA replication and repair, inducing apoptosis and cytotoxicity in FRα-positive tumor cells. This mechanism provides both direct cytotoxicity and bystander killing effects, making GenSci140 promising in addressing tumor heterogeneity and resistance[1][2][3][4][5][7][12].
