Overview

Gentamicin bone cement, developed by Sanskruti Pharma, is a drug-device combination used in orthopedic surgery to secure prosthetic implants and provide local antibiotic prophylaxis. It consists of an acrylic polymer, polymethylmethacrylate (PMMA), which acts as a structural grout, and the aminoglycoside antibiotic gentamicin. Upon implantation, the cement hardens and slowly elutes gentamicin into the surrounding tissue and bone. This local delivery achieves high bactericidal concentrations at the surgical site, effectively targeting common pathogens like Staphylococcus aureus while minimizing systemic exposure and associated side effects such as nephrotoxicity. Gentamicin's mechanism involves binding to the 30S ribosomal subunit of bacteria, leading to misreading of mRNA and inhibition of protein synthesis, which ultimately results in bacterial cell death.
