Genz-644470 is a nonabsorbed, structurally optimized phosphate binding polymer developed for the treatment of hyperphosphataemia in patients with chronic kidney disease (CKD) on hemodialysis. It was designed as a new physical form of sevelamer carbonate, featuring large aggregates of smaller particles to enhance selective phosphate binding and deter entry of competing ions. In clinical studies, Genz-644470 demonstrated effective reduction of serum phosphorus levels in hyperphosphatemic CKD patients on hemodialysis and showed higher phosphate-binding capacity than sevelamer carbonate in vitro. However, its development was discontinued after phase II trials due to lack of significant advantages over existing therapies[1][3][6].
