Drug intelligence / Profile preview
GF9-LPC is a first-in-class TREM-1 inhibitory peptide (GF9) formulated into self-assembling, brain-permeable, macrophage-specific lipopeptide complexes (LPC). Developed by SignaBlok, it employs a ligand-independent mechanism to block Triggering Receptor Expressed on Myeloid cells 1 (TREM-1), an inflammation amplifier. By inhibiting TREM-1, the drug reduces intratumoral macrophage infiltration and lowers serum levels of pro-inflammatory cytokines such as IL-1α, IL-6, and macrophage colony-stimulating factor (M-CSF). Preclinical studies have demonstrated antitumor activity in non-small cell lung cancer (NSCLC), pancreatic cancer, and glioblastoma multiforme (GBM), particularly when used in combination with chemotherapy or radiation. The LPC formulation is specifically designed to extend the peptide's half-life and enable crossing of the blood-brain barrier.
